    Enzymatic Characterization of Leishmania major Phosphatidylethanolamine Methyltransferases LmjPEM1 and LmjPEM2

    Phosphatidylcholine (PC) is the most abundant phospholipid in the membranes of the human parasite Leishmania. It is synthesized via two metabolic routes, the de novo pathway that starts with the uptake of choline, and the threefold methylation of phosphatidylethanolamine. Choline was shown to be dispensable for Leishmania; thus, the methylation pathway likely represents the primary route for PC production. Here, we have identified and characterized two phosphatidylethanolamine methyltransferases, LmjPEM1and LmjPEM2. Both enzymes are expressed in promastigotes as well as in the vertebrate form amastigotes, suggesting that these methyltransferases are important for the development of the parasite throughout its life cycle. Heterologous expression in yeast has demonstrated that LmjPEM1 and LmjPEM2 complement the choline auxotrophy phenotype of a yeast double null mutant lacking phosphatidylethanolamine methyltransferase activity. LmjPEM1 catalyzes the first, and to a lesser extent, the second methylation reaction. In contrast, LmjPEM2 has the capacity to add the second and third methyl group onto phosphatidylethanolamine to yield (lyso)PC; it can also add the first methyl group, albeit with very low efficiency

    Challenges in control and autonomy of unmanned aerial-aquatic vehicles

    Autonomous aquatic vehicles capable of flight can deploy more rapidly, access remote or constricted areas, overfly obstacles and transition easily between distinct bodies of water. This new class of vehicles can be referred as Unmanned Aerial-Aquatic Vehicles (UAAVs), and is capable of reaching distant locations rapidly, conducting measurements and returning to base. This greatly improves upon current solutions, which often involve integrating different types of vehicles (e.g. vessels releasing underwater vehicles), or rely on manpower (e.g. sensors dropped manually from ships). Thanks to recent research efforts, UAAVs are becoming more sophisticated and robust. Nonetheless numerous challenges remain to be addressed, and particularly dedicated control and sensing solutions are still scarce. This paper discusses challenges and opportunities in UAAV control, sensing and actuation. Following a brief overview of the state of the art, we elaborate on the requirements and challenges for the main types of robots and missions proposed in the literature to date, and highlight existing solutions where available. The concise but wide-ranging overview provided will constitute a useful starting point for researchers undertaking UAAV control work

    Assessing the Accuracy of Discourse Connective Translations: Validation of an Automatic Metric

    Automatic metrics for the evaluation of machine translation (MT) compute scores that characterize globally certain aspects of MT quality such as adequacy and fluency. This paper introduces a reference-based metric that is focused on a particular class of function words, namely discourse connectives, of particular importance for text structuring, and rather challenging for MT. To measure the accuracy of connective translation (ACT), the metric relies on automatic word-level alignment between a source sentence and respectively the reference and candidate translations, along with other heuristics for comparing translations of discourse connectives. Using a dictionary of equivalents, the translations are scored automatically, or, for better precision, semi-automatically. The precision of the ACT metric is assessed by human judges on sample data for English/French and English/Arabic translations: the ACT scores are on average within 2% of human scores. The ACT metric is then applied to several commercial and research MT systems, providing an assessment of their performance on discourse connectives

    Systematic inference and comparison of multi-scale chromatin sub-compartments connects spatial organization to cell phenotypes.

    Chromatin compartmentalization reflects biological activity. However, inference of chromatin sub-compartments and compartment domains from chromosome conformation capture (Hi-C) experiments is limited by data resolution. As a result, these have been characterized only in a few cell types and systematic comparisons across multiple tissues and conditions are missing. Here, we present Calder, an algorithmic approach that enables the identification of multi-scale sub-compartments at variable data resolution. Calder allows to infer and compare chromatin sub-compartments and compartment domains in >100 cell lines. Our results reveal sub-compartments enriched for poised chromatin states and undergoing spatial repositioning during lineage differentiation and oncogenic transformation

    Carbohydrate reserves in grapevine (Vitis vinifera L. 'Chasselas'): the influence of the leaf to fruit ratio

    Seasonal dynamics of total non-structural carbohydrates (TNC) in relation to the leaf-fruit ratio were measured over five years at different grapevine phenological stages in one- and two-year-old canes, trunks and roots of the cultivar 'Chasselas' (Vitis vinifera L.). Carbohydrates were mainly stored as starch in different parts of the grapevine during the growing season. Soluble carbohydrates represented only a small part (< 7 % of dry weight, DW) of the TNC. In the roots and trunks, the starch content fluctuated during the growing season, reaching the lowest values between budbreak and flowering depending on the year, and the highest values between harvest and leaf fall. The soluble sugar content increased in the trunks and the two-year-old canes during the winter period with the decrease in temperatures. A negative correlation was established between the average air temperature recorded during the seven days before sample collection for carbohydrate analysis, and soluble carbohydrate content in the trunks and two-year-old canes. The leaffruit ratio (source-sink), expressed by the “light-exposed leaf area∙kg-1 fruit”, not only substantially influenced the soluble sugar content in berries but also the starch and TNC concentrations in the trunks and roots at harvest. Higher leaf-fruit ratios resulted in increased starch and TNC concentrations in the trunks and roots, which attained the maximum values when the leaf-fruit ratio neared 2.0 m2 of light-exposed leaf area∙kg-1 fruit. Canopy height and leaf area had no predominant influence on the soluble sugars, starch contents, or TNC in the permanent vine parts.

    Best Paper Honorable Mention awardInternational audienceWe reflect upon the design of a paper-based tangible interactive space to support air traffic control. We have observed, studied, prototyped and discussed with controllers a new mixed interaction system based on Anoto, video projection, and tracking. Starting from the understanding of the benefits of tangible paper strips, our goal is to study how mixed physical and virtual augmented data can support the controllers' mental work. The context of the activity led us to depart from models that are proposed in tangible interfaces research where coherence is based on how physical objects are representative of virtual objects. We propose a new account of coherence in a mixed interaction system that integrates externalization mechanisms. We found that physical objects play two roles: they act both as representation of mental objects and as tangible artifacts for interacting with augmented features. We observed that virtual objects represent physical ones, and not the reverse, and, being virtual representations of physical objects, should seamlessly converge with the cognitive role of the physical object. Finally, we show how coherence is achieved by providing a seamless interactive space

    The N-Terminal Domain and Glycosomal Localization of Leishmania Initial Acyltransferase LmDAT Are Important for Lipophosphoglycan Synthesis

    Ether glycerolipids of Leishmania major are important membrane components as well as building blocks of various virulence factors. In L. major, the first enzyme of the ether glycerolipid biosynthetic pathway, LmDAT, is an unusual, glycosomal dihydroxyacetonephosphate acyltransferase important for parasite's growth and survival during the stationary phase, synthesis of ether lipids, and virulence. The present work extends our knowledge of this important biosynthetic enzyme in parasite biology. Site-directed mutagenesis of LmDAT demonstrated that an active enzyme was critical for normal growth and survival during the stationary phase. Deletion analyses showed that the large N-terminal extension of this initial acyltransferase may be important for its stability or activity. Further, abrogation of the C-terminal glycosomal targeting signal sequence of LmDAT led to extraglycosomal localization, did not impair its enzymatic activity but affected synthesis of the ether glycerolipid-based virulence factor lipophosphoglycan. In addition, expression of this recombinant form of LmDAT in a null mutant of LmDAT did not restore normal growth and survival during the stationary phase. These results emphasize the importance of this enzyme's compartmentalization in the glycosome for the generation of lipophosphoglycan and parasite's biology

    In the growing field of collective robotics, spatial co-ordination between robots is often critical and usually achieved via local relative positioning sensors. We believe that range and bearing sensing, based on infrared technology, has the potential to fulfil the strict requirements of real-world collective robots. These requirements include: small size, light weight, large range, high refresh rate, immunity against tilting and misalignment, immunity against ambient light changes, and good range and bearing accuracy. Currently, there are no range and bearing systems that have been designed to cope with such strict requirements. This paper presents a custom range and bearing system, based on a novel cascaded filtering technology, complemented by hybrid infrared/Radio Frequency (RF) communication, which has been designed specifically to meet all these expectations.
